3.2.2. Working with a template

To input data into a template, a calibration line is created with a dose (amount) or concentration on the X-axis and a response, which is the measurement of something, on the Y-axis.

A blank, which is the response of the dilution buffer or the matrix where a substance is measured, is essential for logit regression. The blank's concentration is 0 (zero).

Doses/concentrations for the calibration line are input into the yellow "concentration matrix" field as bold numbers. Sample names, either as text or numbers (not bold), are entered into the other fields. Corresponding dilutions are entered in the green "Dilution matrix" area.

Measured absorbances are recorded in the blue "Absorbance matrix" area.

A completed template will resemble the one below. The standard and sample positions can be randomly placed on the plate, provided that the name and dilution position match the measured absorbance.

By clicking the "Get ELISA data" button on the "Logit" TAB sheet, the entries in the three matrices—extinctions, names, and dilutions—are compiled into a list.
