3.1. General instructions

The program can be used with the Excel versions 2002 up to and including 2019.

The program can be opened in the usual way as a worksheet in Excel.

The various functionalities are started by means of buttons.

General The Microsoft Excel ELISA logit program is primarily developed to calculate the concentrations of multiple samples of which the responses are offered in an ELISA format (8 * 12 matrix) by means of a logit regression. The data is copied from the templates in the Data sheet in the Logit sheet, and the sample names, measured values and applied dilutions can be grouped by sample name. The worksheet is also suitable for manually entering the data as a list in the Logit sheet. Both input methods are described.

Macro settings Macro security should be set to low to permit the running of all macros.
However, it is advisable to increase the security level when opening an Excel worksheet from an unknown source.

Calculations and formulas The worksheets are devoid of any formulas within the cells.
Calculations within the application are executed through password-protected "Visual Basic for Applications" (VBA) modules.
Additionally, the data input into the worksheet for graphing or result computation is handled via VBA.
This safeguards against unintended alterations to formulas.
While the program is quite resilient to the deletion of cell contents, the removal of columns and rows may lead to malfunctions and error messages.

Free note space The program erases parts of the sheet during calculations. An empty space for notes, such as used lot numbers and information, can be used from line 53 in columns H through Q.

Number range Using numbers between 1 and 1000 is preferable for optimal graph presentation.
For example, typing '234' for a milli-extinction is quicker than '0.234'.
Select concentrations for the calibration curve that yield results within the 1 to 1000 range.
For instance, use 10 µg/L instead of 0.00010 g/L, or 20 km instead of 20,000 meters to avoid reading errors.

General Advice Create a template indicating the positions of standards, samples, and their respective dilutions.

Save the template with a unique identifier plus the original file name to preserve the version number.

Set this file to Read-only to avoid accidental overwriting. Validate the template as needed.

Employ this blank template for each new test and refrain from reusing old worksheets.
